新規薬物依存タンパクshatiの生理機能解析

Research Abstract

薬物依存の形成に寄与すると考えられるいくつかの遺伝子が報告されている。薬物依存の形成は多くのファクターが複雑に絡み合い、未知の遺伝子が関与している可能性も高い。そこで、我々は、覚せい剤であるメタンフェタミンを連続投与したマウス脳において発現量の高い2つの遺伝子をcDNAサブトラクション法を用いて見出した。cDNA断片から全長をクローニングしたところ、1つは、すでに既知のタンパクでありpiccoloと命名され、インシュリン分泌に関係するタンパクとして研究がなされていたが、脳での生理機能については、ほとんど分かっていなかった。もう一方は、新規の遺伝子であったことから、shatiと命名し、全長の単離・同定に成功した。両タンパクの遺伝子配列にもとづいて作成したアンチセンスヌクレオチドを脳に注入し、各タンパクの発現を減少させたマウスでは、覚せい剤による場所嗜好性や自発運動の亢進が促進されたことから、両遺伝子は薬物依存形成を抑制すると考えられる。In vivoマイクロダイアリシス法による実験では、これらマウスにメタンフェタミンを7-10日間連続投与し、最終投与後のドパミン遊離量を測定したところ、遊離量増大が促進された。これらの結果から、両遺伝子の薬物依存形成抑制作用は、ドパミン遊離の調節を受けていると考えられた。さらに培養細胞やex vivoの実験によって、両遺伝子が、ドパミントランスポーターの機能調節に関係しているという新しい知見を得た。次年度以降は、両遺伝子の組み換えマウスの作成を行い、生理機能の解明を行う予定である。
