植物における免疫活性化機構と病原体による免疫抑制化機構の解明

Research Abstract

本年度は本研究室において同定された植物免疫シグナル伝達系における重要タンパク質、RAR1,SGT1,HSP90,U-Boxタンパク質群、MAPキナーゼカスケードタンパク質群などを中心にその構造解析を進めた。候補タンパク質の大量精製については可溶性の高いものから随時、ミニスケールで大腸菌での発現をテストした。いくつかのタグシステムを試し、発現量が高く、可溶各分にいくものから大量発現および精製の対象とした。これによって発現が難しいものは、無細胞系翻訳システム(大腸菌ベース、小麦胚芽ベース)を用いた。大量精製が可能なタンパク質あるいは複合体は白水美香子博士の研究室において随時結晶解析を行った。構造解析の成否はターゲットとなるタンパク質の可溶性などの性質に大きく左右されるため、数多くのターゲットタンパク質の可溶性をハイスループットシステムを使ってスクリーニングしてその成功率を高めた。構造が決定したものについては、アミノ酸置換を導入して機能解析を行った。例えば、複合体の構造解析から示唆される結合領域にあるアミノ酸を置換し、相互作用に影響があるかをテストし、結晶構造の生物学的意味を検討した。また植物病原体由来のエフェクタータンパク質群についても構造解析を推し進めた。NMRを用いて解析したものはFOLDINGがうまくいっていないNATURAL DISORDERED PROTEINであることが判明した。
