Research Abstract

精子幹細胞は成体の精巣にあり、一生にわたって精子形成の源となる細胞である。研究代表者らは2003年にマウスの精子幹細胞の長期培養系を確立し、これをGermline stem(GS)細胞と命名した。この細胞の特徴は非常な長期にわたり安定的に増殖することであり、2年間で10^<85>倍(139 passages)に増え、その間増殖速度や幹細胞としての活性×インプリンティングや核型などに全く変化を示さず、2年の培養後も正常な子孫を作った。
本研究の目的は幹細胞の安定した自己複製機構がどのようなメカニズムで成り立っているのか、そしてその破綻がどのように起こるのかを精子幹細胞を使って明らかにすることである。具体的には、GS細胞の2年以降の増殖を追跡し、寿命の測定とともに、どのように終焉を迎えるかを観察する。またGS細胞の自己複製を司るシグナル伝達経路の解明と、寿命と自己複製活性の関連について調べる。
