キネシンとダイニンを用いた分子ナノソーティングシステムの開発

Research Abstract

本研究の全体構想は、生体内物質輸送を担っているモータタンパク質の駆動系をマイクロ・ナノデバイス内で再構築することによって、キネシンとダイニンの運動によりナノスケールの対象分子などを大量・並列に操作できるナノアクチュエータを創製することである。平成19年度において、キネシンとダイニンを付加したビーズの同時搬送と微小流体デバイスめ製作をおこなった。本年度は、それらの要素技術を用いて分子ソーテイングを実現するため、以下の二項目について研究を実施した。
1. 目的分子結合方法の検討
本項の目的は、アビジンービオチン結合などの分子特異的な結合を用いて、ビーズ表面に目的分子を固定する技術を開発することである。目的分子として、ビオチン(biotin-4-fluorescein)とIgG(Rabbit anti-mouse IgG AlexaFluor 568)のビーズへの付加を確認することができた。特に、目的分子の濃度によってキネシンおよびダイニンの活性が変化することが分かった。
2. 分子ソーティングの実現
マイクロ流路内に配向した微小管上で、分子ソーティング技術の検討をおこなうことを目指した。微小管の配向、各ビーズの搬送を確認することができた。ただ、ビオチンやIgGを付加したビーズを大量かつ同時に搬送、ソーティングには至らなかった。今後、定量的に目的分子の付着量を評価し、ビーズの同時駆動を実現させたい。
