三次元培養肝細胞を利用した肝炎ウイルス感染モデルの開発

Research Abstract

平成19年度にクローニングしたHBVをタンデムに結合して作成したウイルス発現用ベタターをヒト肝癌由来細胞株Huh7細胞に導入し、この培養上清を超遠心操作により濃縮した。平皿培養もしくはあらかじめメビオールゲルにて三次元培養しておいたnaiveなHuh7細胞、Alexander細胞とincubateしたのち、数日間培養を継続し経時的に細胞を回収した。その結果、三次元及び平皿培養細胞どちらも感染後HBsAg量はnegativeにはならないものの経時的に減少した。
ウイルス感受性の有為な差が得られなかったことから、ラジアルフロー型バイオリアクター(RFB)の系による感染実験を試みた。本装置は人工臓器として開発されたものであり、遺伝子型によらないHCV患者血清の感染および複製が観察できる系である。RFBでFLC4細胞を培養し、HBV感染後中和抗体が出現する前の急性期の患者血清を用いて感染実験を行った。その結果、感染後HBsAgは陰性になったが、約1ヶ月後から弱陽性となった。
本研究により、三次元培養がHBV感染の感受性を変化させる可能性が示唆された。研究代表者は、他の研究成果から、いくつかの細胞接着因子は細胞の三次元によって細胞質から細胞接着面にその局在をかえる事を見いだしている。この事からも、ウイルスのレセプターが三次元化により細胞表面に局在出来るようになった、または、必要なcomnlexを形成できるようになったことが考えられる。今後、細胞膜表面蛋白のプロテオーム解析等によってHBVの新規受容体が同定できる可能性があるだろう。また、培養細胞を用いたHBV長期感染系は報告されていない。本研究の成果は、薬剤耐性ウイルスの解析等にRFBが利用できる可能性を示唆した。また、簡易的な三次元培養系であるメビオールゲルと比べ、RFBがより生体に近い環境を作り出せる事をウイルス感受性の違いで示すことが出来た。今後人工肝臓の機能評価においてHCV, HBVウイルス感受性が指標として利用できる可能性も考えている。
