動脈硬化プラーク破綻におけるマクロファージ・アポトーシスの役割解明と治療法

Research Abstract

1. プラーク破綻モデルマウスの作成
本研究において8週間の高脂肪食を負荷したApoE欠損マウスに4週間アンギオテンシンII持続負荷を行うことにより、腕頭動脈に高頻度の急性プラーク破綻(64%, n=11) およびburied fibrous cap(2.8±0.4/個体, n=11)生じるマウスモデルを作成した。
2. マクロファージ選択的ナノ粒子の作成
マクロファージ選択的に治療因子を導入するため、径200nmのpoly(lactic-co-glycolic acid)(PLGA)ナノ粒子を作成した。FITCを封入したPLGAナノ粒子(FITC-NP)をマウスへ静脈注射したところ、1時間後の末梢血ではCD11b(+)Gr-1(-)の単球に選択的にFITCの取り込みを認めた。また、高脂肪食負荷を行ったApoE欠損マウスにFITC-NPを静脈注射したところ、腕頭動脈・動脈硬化巣内のマクロファージにFITC免疫染色を認め、PLGAナノ粒子が単球、マクロファージ選択的キャリアとなることが示された。
3. PPAR-γアゴニストであるピオグリタゾンは、マクロファージにおいてアポトーシスを誘導することが示されている(Thorp, Circulation 2007)。ピオグリタゾンをPLGAナノ粒子に封入し、上記プラーク破綻モデルマウスに対する静脈内投与を行ったところ、4週間後のプラーク破綻の減少を認めた。
4. 今後、治療効果の機序についての検討を進める予定である。
