Research Abstract

乳児急性リンパ性白血病において最も多く見られる、MLL-AF4ファミリー融合蛋白の白血病発症機序についてレトロウイルス遺伝子発現系を用い解析した。本研究ではMLL-AF4, MLL-AF5q31について機能解析を行い、AF4ファミリー蛋白に共通して保存されているC-terminal homology domain (CHD)がマウス造血幹細胞の不死化に必須の領域である事を明らかにした。また、本領域の機能としては、AF4CHDとAF5q31CHD間の結合能を明らかにした。AF4, AF5q31はENL, hDot1Lと結合し、H3K79dimethyltransferase活性をもつ、蛋白複合体を形成する事が知られており、白血病発症との関わりについて、次期研究で検討中である。
