| Research Abstract |
ヒト骨肉腫細胞株(OS)を用いIL-18がOSのFasL抵抗性に及ぼす影響を検討した。4種のヒト骨肉腫細胞株はIL-18receptorを発現しているが、腫瘍増殖、Fasの発現には全く影響をおよぼさず、遊離型Fas(sFas)の分泌においても変化はみられなかった。次にhistone deacetylase inhibitorであるsodium valproate(VPA)について上記と同様の目的にて検索した結果、VPAはヒト骨肉腫細胞の増殖を抑制した。またsFasの分泌を抑制し、FasLに対する感受性を増加させ、OSの免疫細胞抵抗性を減弱させることが示された。さらにVPAの前処置はFasを介したapoptosisのsignal伝達を活性化させることが示された。VPAはすでに抗てんかん薬として現在広く臨床応用されており、安全性・至適濃度はすでに確認されている。今回使用した濃度はその範囲内であることから、人体への影響が少なく、骨肉腫の治療に応用可能であることが示された。
