Research Abstract

ヒト抹消血単球細胞THP-1細胞由来マクロファージにおいて、IL-1β依存的ケモカイン産生に対してPGE2の作用を検討した結果、IL-8, RANTES, MIP-1β,MCP-3の産生が、PGE2添加により有意に抑制することが確認された。また、THP-1細胞におけるEP受容体の発現を確認した結果、EP1,2,3,4は恒常的に発現していることが確認された。さらに、EP2およびEP4アンタゴニストを用いてIL-1依存的IL-8産生におけるPGE2の作用を検討した結果、EP4アンタゴニストはPGE2により抑制されたIL-8産生を有意に増加させたが、EP2アンタゴニストは影響を及ぼさなかった。
今回の研究よりPGE2の抗炎症作用にはEP4受容体が関係していることが確認された。
