Outline of Annual Research Achievements

The system pulse-shaping capabilities were improved and a transform-limited pulse was achieved, improving the sensitivity and signal quality for carbohydrates and carotenoids. Heavy ions mutagenesis was performed in Euglena gracilis, identifying hyperaccumulating mutants via FT-CARS paramylon. RNA isolation was done and cDNA analysis was performed. Analysis of this data and manuscript preparation is currently in process. The analysis of starch accumulation in Chromochloris zofingiensis via both FT-CARS and enzymatic analysis were done, and a manuscript is in preparation. Sorting of multiple microalgae species was achieved and findings were recently published in Advanced Materials Technology. Human blood signal was obtained via FT-CARS, likely due to the strong resonant Raman of haemoglobin. However, leukocyte detection is still challenging with the current system sensitivity, and thus the system will require further optimization for clinical applications. An additional application of the FT-CARS system was identified: microplastics detection under flow. Preliminary work was done and has been followed by other researchers after the fellow’s departure.
