Outline of Annual Research Achievements

鏡像体アラビナン分解酵素のうち、2種類のEndo型酵素(EndoMA1, EndoMA2)と2種類のExo型酵素(ExoMA1, ExoMA2)の機能解析と構造解析を進めた。さらに、EndoMA1の結晶構造で発見した新規CBMの金属ナノ粒子(GNP)を用いた解析を進めた。 EndoMA1とEndoMA2の基質特異性解析を合成基質を用いて行った。EndoMA1の新規CBMのGNP解析では、22-30糖のD-アラビナン(D-Ara22-30)、9糖の直鎖オリゴ糖(A9L)、7糖のβ結合を有する分岐オリゴ糖(A7BβT)、5糖の直鎖オリゴ糖(L-Ara5)を固定化したDNPに対し、野生型酵素、活性中心変異体(D51N, D33N)、新規CBM結合部位変異体(F379A, W468A)、新規CBMを欠失したコンストラクト(C-del)などを用いて凝集反応を調べた。その結果、凝集には二量体の形成が重要なこと、9糖以上のD-アラビナンオリゴ糖であれば凝集が起こること、新規CBMの欠失および変異体では結合が弱くなることなどが明らかとなった。ExoMA1ではクライオ電子顕微鏡解析による立体構造決定に成功した(分解能2.29オングストローム)。ExoMA2ではD557N, D557A, E431Q, E431Aの合計4種の変異体の機能解析を行い、阻害剤プローブであるDNP 2F-β-D-Arafによる阻害定数を決定した。さらに、ExoMA2の二糖Araf-β1,2-Arafおよび七糖A7BβTとの複合体の構造決定に成功し、サブサイト+1以降の基質認識機構を明らかにした。一方、本研究で使用した基質、リガンドおよび阻害剤プローブの立体選択的糖鎖合成の開発検討を行い、大量に合成して実験に用いた。
