Outline of Annual Research Achievements

本課題では、淡水環境への適応に重要な遺伝子を同定し、当該遺伝子座におけるクロマチン構造上の特徴を見出すことである。本年度は、淡水進出の鍵となる遺伝子について、機能解析のためにノックアウトを実施した。その結果、淡水進出に際して変化する多数の形質(体サイズ、繁殖時期、ホルモン量など)を同時に変える鍵遺伝子であることを明らかにした。この遺伝子の適応度への影響を解析するために、海型と淡水型のF2雑種を作出し、メソコスムに放流し、メソコスムにおける適応度を解析しRAD-seq(Restriction Site Associated DNA Sequence)にてジェノタイピングを実施した。また、さまざまなエコタイプについて、ATACシークエンス(Assay for Transposase-Accessible Chromatin with high-throughput sequencing)と修飾ヒストン抗体てCUT&Tag(Cleavage Under Targets and Tagmentation)-シークエンスを実施した。その結果、エコタイプ間で上記の遺伝子を含むいくつかの領域でクマチン状態が異なることを見出した。また、エコタイプ間でのクロマチンの違いを生み出す遺伝基盤を解明するために、QTL(Quantitative Trait Loci:量的形質遺伝子座)解析を実施した。これらデータをまとめて論文化にもっていける状態になった。
