生体分子の分子認識プロセスの時空間的スケールギャップ問題の解決のための基礎研究

Outline of Annual Research Achievements

当該年度においては、原子間力顕微鏡(atomic force microscopy: AFM)と高速オシロスコープの複合装置を構築し、ストレプトアビジン-ビオチン間に働く、相互作用を高時間分解能で、解析した。2019年にRSC Advancesに発表した観察結果をさらに上回る時間分解能での解析に成功し、さらにはその相互作用力を2次元でマッピングすることで、バイオセンサー上の受容体(抗体)分子のターゲット分子への結合能を定量的に評価する手法を確立し、論文発表(Nanoscale)を行った。
また当該年度に構築した手法は、他のリガンド-受容体、抗原抗体反応の解析にも利用出来ることを報告し、当初の研究目的を達成した。

Reason

当初の目的である、リガンドー受容体間の相互作用の高時間分解能測定を達成し、論文発表まで行ったため、順調に研究が進展していると言える。また、また当該年度に構築した手法は、他のリガンド-受容体、抗原抗体反応の解析にも応用できることを確認出来たことから、確実に研究のターゲットをクリアできる状況にある、と判断した。

Strategy for Future Research Activity

次年度は解析したストレプトアビジン-ビオチンの完全結合・完全解離状態の間に存在する、準安定状態の理論的解析を行う。特に、分子動力学法を用いた分子シミュレーションを組み合わせ、より詳細な分子レベルでの結合のダイナミクスに関して議論する。
