Outline of Annual Research Achievements

本研究では,マイクロ流体工学を用いて簡便かつ高効率にナノ~ピコリットルサイズの巨大脂質膜小胞を区画とした均一なバイオリアクターを製造する方法を確立し,がんなどの疾患バイオマーカの検出に応用することを目的とした.2022年度は,本研究で開発した,マイクロ流路を用いた均一GUV形成法の応用範囲を拡張した.GUV内で無細胞タンパク質合成系が動作する条件は2021年度に確立したので,2022年度は特に膜タンパク質の再構成が行える条件を探索した(論文投稿中).さらに,共同研究において,この技術を応用し,均一GUV内でのDNAゲルの形成過程の解明を行った.これらの技術により,人工細胞外に存在するバイオマーカとしてのmiRNAを内部に取り込んで検出する一連の要素技術を確立することができた.当初の目標であった,増幅反応を用いたmiRNAの検出についても,等温のEXPAR(Exponential Amplification Reaction)反応を用い,顕微鏡およびフローサイトメトリーで蛍光検出する方法を確立した(論文執筆中).膜融合をトリガーとした検出方法を確立することで,疾病診断用途への応用が広がると期待される.また,派生テーマとして,振動誘起流れを用いて微小な流れ場をつくり,極微量サンプル中のmiRNAを免疫凝集法で検出する技術を確立した.以上のように,申請当初に提案した内容を達成するだけでなく,複数の派生テーマが進行し,バイオマーカ検出のレパートリーを増やすことに成功した.
