Outline of Final Research Achievements |
In this study, we established immunoassay methods in aqueous microdroplets using spontaneous emulsification. The first method is a sandwich immunoassay utilizing inorganic salt enrichment by spontaneous emulsification and accumulation at the interface of the nanoparticles. The second method, competitive immunoassay, was established using selective separation of small molecules by spontaneous emulsification. This technique enabled the semiquantitative detection of secretions from multiple cells in a single droplet. In addition, through this study, we found the factors that cause spontaneous emulsification by Span 80: the smaller the Span 80 reverse micelle, the lower the chemical potential of water, and the difference in chemical potential drives spontaneous emulsification.

Academic Significance and Societal Importance of the Research Achievements |
本研究では、自然乳化を利用したマイクロ水滴内イムノアッセイを確立した。特に、粒子を用いた手法では自然乳化による定量的濃縮操作を利用することで、マイクロ流体デバイスを用いることなくBF分離を実現できる可能性がある。本手法により、今後、細胞分泌物のハイスループットな検出とそれに基づく細胞分離が可能になると期待する。 また、Span 80を用いた自然乳化メカニズムを明らかにした。自然乳化は外力不要乳化法であり、エマルションのみならずナノ粒子やドラッグキャリアなどの材料合成にも応用されている。本成果を通じ、自然乳化を起こしうる新たな界面活性剤のデザインが可能になり、産業応用の拡大につながると期待する。
