Outline of Annual Research Achievements |
All range-wide DNA samples of the subalpine trees Tsuga diversifolia, Thuja standishii, Picea jezoensis and Pinus koraiensis have been extracted and RAD-seq data for both Tsuga diversifolia and Thuja standishii has been obtained. Sampling of the other target species of this project (Abies vietchii, Ilex rugosa and Rhododendron brachycarpum, Betula ermanii and Vaccinium vitis-idaea) has began with first collections from the southern range limit of Mt Ishizuchi, Ehime Prefecture. For Thuja standishii and Tsuga diversifolia, whole chloroplast genome data has been obtained and genetic differences between southern edge and the range-centre identified.

Reason
Collections have been successful in locating the target species and I expect that most sampling will be completed this financial year. DNA extraction methods for each species have been optimised and the RAD-seq and whole chloroplast DNA data for most species should be completed this financial year.

Strategy for Future Research Activity |
This financial year I plan to complete collections from remaining uncollected southern edge populations in Kyushu, Shikoku and the Kii Peninsula and to sample representative sites in other regions of Japan. I will also try to obtain samples from Korea and Far East Russia for the species that occur there. DNA will be extracted and sent for RAD-sequencing. Chloroplast DNA variation will also be searched for using whole chloroplast genome sequencing.
