Outline of Final Research Achievements |
The aim of the project was to characterize enzymes involved in biosynthesis of plant cell wall pectin. In this study, glycosyltransferases and acetyltransferases involved in pectin biosynthesis were characterized. The galacturonosyltransferases involved in the synthesis of the pectic rhamnogalacturonan I (RG-I) were found to have eight genes in the Arabidopsis genome. The galactoryltransferase involved in synthesis of RG-I side chain was detected for the first time and given the enzyme number EC 2.4.1.375. The enzymatic activity of RG-I: acetyltransferase was detected for the first time in TBL7. In addition, a substrate for RG-II: Kdo transferase, CMP-Kdo, was prepared.

Academic Significance and Societal Importance of the Research Achievements |
植物細胞壁ペクチンのいくつかの生合成酵素を解析・遺伝子同定した本研究成果は、ペクチンの機能解析に役立てられる。ペクチンの機能として考えられている細胞接着・形態形成・硬軟調節などに焦点を当て、本研究で同定したペクチン生合成酵素遺伝子を用いて、機能解析を進めることができるようになる。また、ペクチンがヒトの免疫機能を活性化すると考えられており、本研究成果により、ペクチンのどの構造がその機能に関与するか明らかにする研究を展開できる。
