細胞活性化時の生理機能変化を担う「分子レバレッジ機構」の解明

GPIIb/IIIaの細胞内・膜貫通・細胞外ドメインの連成モデルをXeon PhiおよびGPU 4基を実装した学内コンピューターに導入した。初期構造からエネルギー的安定構造探索を目指す計算を継続した。水分子を除く全ての原子の距離の2乗平均値（root mean square deviations: RMSDs）を構造安定の指標とした。
2021年度にはGPIIb/IIIaの細胞内・膜貫通・細胞外ドメインのRMSDと、GPIIIb/IIIaを構成するGPIIb、GPIIIa、両分子複合体の細胞外、膜貫通、細胞内ドメインのRMSDを計算した。全体のRMSDが収束した条件にて、GPIIIaの構造はRMSD 8オングストローム程度に収束したのち再度不安定化した。GPIIbは859のアミノ酸から構成されるheavy chainと、107のアミノ酸から構成されるlight chainからなる。特にlight chainの構造の不安定性が示唆された。
GPIIbの細胞内、膜貫通、細胞外の各ドメインに注目すると、細胞内ドメインはRMSD 3オングストローム程度に収束した。膜貫通ドメインは細胞内ドメインよりは揺らぎが大きかった。しかし、4オングストローム程度に収束した。細胞外ドメインはheavy chain, light chainともにRMSDは8オングストローム程度に収束する傾向を呈したのちに10オングストローム変動し安定を確認できていない。エネルギー的安定構造の探索計算の継続により細胞内ドメインおよび膜貫通ドメインの安定構造は確定できた。しかし、細胞外ドメインの構造は不安定であり、不安定性は主にGPIIbの細胞外ドメインの構造によることが2021年度の研究により明らかになった。

本研究ではGPIIbとGPIIIaの2つの膜糖蛋白が複合して血小板上のフィブリノーゲン、von Willebrand因子などの活性化依存性の接着を担う膜糖蛋白GPIIb/IIIaの細胞外ドメインの活性化構造変化を惹起する「分子レバレッジ」の解明を目指す。細胞外ドメインは結晶構造を適応した。しかし、膜貫通ドメイン、細胞内ドメインは任意に作成する必要があった。2021年までの計算によりGPIIb/IIIaの細胞内・膜貫通・細胞外ドメインの一体的計算により細胞内ドメインおよび膜貫通ドメインの構造の安定化を確認できた。細胞外ドメインは完全な安定に至っていないが、結晶構造と大きな差がないことを確認できた。
各ドメインの安定構造解析により、GPIIbの細胞外ドメインに構造の不安定性が集中していることがわかった。分子レバレッジにはGPIIb/IIIaの細胞外ドメインの役割が大きいことが示唆された。各ドメインの構造の安定性の定量評価であるRMSDの計算も着実に進行した。
GPIIb/IIIaの高次構造の動的解析には「京」、「富岳」などの学外コンピューターの使用が必須と当初想定されていた。しかし、GPUを実装した学内コンピューターにより、必ずしも学外コンピューターの使用は必要なくなった。GPIIb/IIIaのエネルギー的安定構造を確定したのちに細胞内ドメインを拘束することが当初の予定であった。GPIIbの細胞外ドメインの不安定性が明らかになったので、当初の予定通りに細胞内ドメインを拘束する研究と並行して初期構造からの構造計算も必須と考えている。研究は概ね順調に施行できているが、今後の結果に注視する必要がある。

2021年までの研究により、GPIIb/IIIaの細胞内・膜貫通・細胞外ドメインの動的構造解析計算は順調に進んでいる。しかし、GPIIbの細胞外ドメインの構造の不安定性が確認されたため、当初の予定通りにGPIIb/IIIa全体のエネルギー的最安定構造自体が不安定である可能性がある。当初の予定通りに、細胞内ドメインを拘束し、細胞外ドメインの構造変化を計算する研究と並行して、初期構造からの計算も今後継続したいと考えている。研究開始当初よりも学内計算資源が充実したため、2方向の計算を並列的に施行することは可能と判断している。今後、途中経過として算出される構造の変化を注視し、計算資源の適切な配分を決めて行きたい。
