Outline of Final Research Achievements

In this study, we examined whether salivary gland stem cells were located at the top of the hierarchy of salivary gland cell lineages. We labeled submandibular gland cells with random color using Rosa26-CreERT2/rainbow mice and pursued cell differentiation. Consequently, bipotent stem cells existed on embryonic day 13.5, while only unipotent stem cells existed after embryonic day 16. Therefore, these results suggest that tripotent stem cells possibly exist in submandibular glands before embryonic day 13.5.

Academic Significance and Societal Importance of the Research Achievements

唾液腺は、導管上皮細胞、腺房細胞および筋上皮細胞の3系統の細胞よりなり、これまで介在部導管に存在する多分化能を有する幹細胞により３系統すべての細胞供給がなされていると考えられてきた。しかしながら、成獣マウス唾液腺においては、分化した細胞が個別に細胞を供給していることが明らかとなり多分化能を有する幹細胞は存在しないことが報告された。一方、本研究では、マウス胎生期唾液腺において幹細胞が段階的に多分化能を消失することが示された。本研究により得られた結果は、唾液腺障害に対する細胞治療の移入ソースの開発に極めて有用な情報となると考えられる。
