Outline of Annual Research Achievements

1. 特異的味成分単回添加後の脊髄後根神経節における神経活動亢進の検証(ex vivo解析)
カルシウムバイオセンサーYC3.60トランスジェニックマウスから、消化管感覚神経の細胞体が存在する脊髄後根神経節から神経細胞を単離し、初代培養細胞系を確立した。この評価系にモデル化合物である(-)-epicatechin四量体・cinnamtannin A2(A2)を添加したところ、陽性対象であるcapsaicinおよびアリルイソチオシアネートと同様に顕著なCaシグナルが観察された。
2.特異的味成分投与直後のニューロトランスミッターおよび関連遺伝子変動の解析
野生型マウスにA2を強制経口投与した直後に断頭し、ノルアドレナリンシグナルを質量MSイメージングにて解析した。その結果、青斑核・視床下部・脳幹に顕著なシグナルが観察された。また同時にin situ hybrdization法を用いて、ノルアドレナリン生成酵素およびトランスポーターの遺伝子発現変動を観察したところ、青斑核にのみ上昇が観察された。このことから、A2の摂食刺激は、青斑核に入力され、視床下部および脳幹に投射されることが明らかとなった。本研究成果は2019年に行われたInternational Conference on Food Factors2019および2020年度日本農芸化学会大会で発表した。

2019年度の研究成果は、上記に示したとおりであり、提出した研究計画どおりに進んでいる。

Strategy for Future Research Activity

当初計画では、特異的味成分反復投与後のストレス耐性獲得作用の検証および特異的味成分反復投与後の自律神経調整を介した循環・代謝亢進作用の確認については2021年度に実施する予定であったが、脳内神経伝達物質の挙動と合わせて解析することが望ましいと考えることから、前倒しして2020年度より着手することとする。
