双生児のエピゲノム情報と臨床検査値に基づいた、生活習慣病に影響する環境因子の解明

| Outline of Annual Research Achievements |
① 表現型不一致双生児・一致双生児の同定（並びに継続サンプリング）：臨床検査値を表現型として、一卵性双生児の継続サンプリングを行い、生体試料と疫学データをさらに蓄積した。大阪大学ツインリサーチセンターにおえるサンプリングと併せて、双生児サンプルは600組1200名を超え、経時的なサンプリング実績も80組程度となった。また臨床検査値の標準化を行うとともに、タイピングしたSNPデータのQuality ControlおよびImputationを行い、タイピングキットのロット間差等のコントロールを引き続き行った。さらに、DNAメチル化アレイデータのQuality Controlも行うとともにアノテーション情報を追加した。
② 不一致双生児特異的な遺伝背景の同定：既報の臨床検査値の遺伝因子モデルを用いて、過年度では行わなかった、生活習慣病に関係する臨床検査値のGeneticRiskScoreをさらに算出し、臨床検査値への遺伝要因寄与率を反映するものとして個体ごとに同定を終えた。
③ エピゲノム情報（DNAメチル化）の比較：②の臨床検査値の実測値と予測値の乖離を調べ、それぞれの個体における環境要因の寄与度を解析した。また生活習慣の指標となる疫学情報との関係を解析した。
④一卵性双生児ペアごとのメチル化率の差、およびその経年変化を網羅的に解析し、日本臨床検査医学会、日本双生児学会、日本エピジェネティクス研究会で概要を発表した。

臨床検査値について遺伝要因寄与度と環境要因寄与度を網羅的に同定でき、エピゲノム変化との関連解析が進行するとともに、メチル化程度の双生児での網羅的解析の概要が完成した。

臨床検査値への環境要因寄与度と双生児のペア内差とDNAメチル化との関連を引き続き解析し、個体の生活習慣等がそれらに影響している可能性を模索するとともに、双生児間でのメチル化率差の分布を基礎的データとして確立させる。
