Outline of Annual Research Achievements

本研究では、超高感度な光干渉法である統計干渉法(SIT)およびバイオスペックル光断層画像法(bOCT)に基づいて,植物の環境に対する極短時間の生理的・形態的応答を多元的に知ることにより,新しい植物の環境ストレスモニタリング技術を確立することを目指した。
SITを用いて得られる秒オーダーの成長挙動をサブナノメータの分解能で連続的に計測できるシステムを用いて,植物表面の2次元的形態応答に見られる新規な現象である植物成長の「自発的ナノメータゆらぎ」(NIF)の特性を利用する.加えて,新たに提案したbOCTを用いた植物内部の3次元的生理応答情報を統合して利用する。バイオスペックルと呼ばれる信号は生体内部の物質の輸送,細胞器官の運動,微細構造の変化を反映する動的な光散乱現象である.これらを総合して植物の環境応答を多元的に無侵襲,迅速かつ高感度に評価するシステムを構築した.
具体的な環境応答の計測例として、酸性鉱山排水(AMD)に対する植物応答を計測した。発芽前の種子に対するAMD(0, 24, 48mg/L)の暴露実験では特にbOCTによる内部活性に大きなストレス反応が観測された。この反応は植物種によって異なるが、暴露開始から1時間(カイワレダイコン)から24時間(イネ)の早期に検出可能であった。一方発芽後では特にSITによるNIFの変化が顕著であり1時間で検出が可能であった。
これらのAMDストレス応答は従来の生化学的測定、具体的には抗酸化酵素活性(SOD, CAT), H2O2, MDA, TTCテストをおこなった。一方、形態的ストレス測定法である、地上部や根茎の長さ、乾燥重量測定を実施することにより検証した。これら従来法は破壊的な手法でありまた、有意な計測結果を得るために7から10日を要した。従って提案手法は無侵襲かつ極早期に環境ストレスを評価することが可能であることが実証された。
