Outline of Final Research Achievements

(1) The usage of a mixture of three agents as anesthesia in a single prolonged stress procedure (sequential exposure to restraint, forced swimming, and agents inducing anesthesia) for a rat post-traumatic stress disorder (PTSD) model was insufficient for PTSD induction compared with the usage of ether. (2) Chronic voluntary oral ingestion of nicotine in rats reduced depression-like anhedonic behaviors, but it did not affect depression-like learned helplessness behaviors. (3) The placebo effect was not detected with the pain agent tramadol using rat operant behaviors. (4) The exposure to a postnatal enrich environment prior to chronic mild stress procedures increased the expression of resilience to depression-like behaviors in rats.

Academic Significance and Societal Importance of the Research Achievements

本課題においては心的外傷後ストレス障害および鬱に着目し、複数の観点から動物実験を行った結果、鬱におけるアンヘドニア(快感消失の状態)の側面にニコチンによる治療効果が示唆されること、エンリッチメント環境における生育がストレス刺激へのレジリエンスを高める可能性があることなどを明らかにした。精神疾患モデル動物の症状緩和につながる手法を探索することは、ヒトにおける新たな治療法の発見への手がかりとなり得る点で意義がある。
