Outline of Final Research Achievements

Intestinal secretory immunoglobulin A (SIgA) is produced by T-dependent (TD) and T-independent (TI) mechanisms and discrimination between TI-SIgA and TD-SIgA is difficult in the same individual. Thus, to estimate the behavior of both SIgA, we used DO11.10 mice which a naive mouse mainly produce TI-IgA and sensitized with oral specific antigen mice produce TD-IgA in serum and feces.
DO11.10 mice fed with non-purified diet or purified diet (PD), and fecal SIgA and serum IgA were compared. TI-SIgA and TD-SIgA drastically decreased by PD administration though serum IgA was little changed. Additionally, we indicated the correlation between that fecal TD-SIgA and serum TD-IgA in orally sensitized DO11.10 mice. Then, we indicated the expression of polymeric immunoglobulin receptor (pIgR) which is major transporter of IgA decreased in PD fed mice. These findings suggested that PD administration suppressed SIgA with inhibition of IgA transport to the intestinal lumen.

Academic Significance and Societal Importance of the Research Achievements

腸管などに代表される粘膜免疫系では、獲得免疫によって産生される抗原特異的IgA(TD-IgA)の他、抗原非特異的IgA(TI-IgA)が恒常的に病原体の侵入を防いでいる。よって、その維持増進は健康に非常に重要と考えられるが、その産生を調節する機構などは未だ不詳である。本研究は食品及びその成分が腸管のTI-SIgAを調節しうることを明らかとし、またその機序の一部を解明した。本成果は健康維持・増進に有益な食品・食事の開発に道を拓くと期待できる。
