Outline of Annual Research Achievements |
In the past one year, we have successfully achieved the following objectives: (1) We completed the Prince Charming (PC) genetic screen and isolated 36 PC mutants. We are currently focusing on 2 PC mutants, namely PC8 and PC36. We are in the process of generating a mapping population for whole genome sequencing. (2) We have generated a series of glycosylation mutant for Sleeping BEauty (SB) through point mutagenesis. We found that mutation of 5 putative glycosylation sites totally abolish the SB function in promoting polarized tip growth. (3) We have identified ARFC2 as a downstream target for the SB-dependent signaling. ARFC2 knock-out mutant generated also show phenotype which is consistent with the SB overexpressor.

So far we have been able to stick to the original proposed plan.

Strategy for Future Research Activity |
In the next fiscal year, we plan to identify the causative mutations of the two selected PC mutants by engaging commercial sequencing services. We also plan to complete the SB interactome.

Causes of Carryover |
Due to some unforeseen circumstances, we are able to make progress in the research plan by spending less budget. However this year we envisage to spend the money fully to achieve even greater progress.
