Outline of Annual Research Achievements

希少がんでは有効な抗がん剤の開発が望まれているが、薬効の評価に必要なモデル系が入手できないことが課題となっている。体外で腫瘍を維持できるバイオリアクターの開発を目標とした。本研究では患者から採取した腫瘍組織をバイオリアクター内で長期間維持し、臨床に近い条件で抗がん剤感受性試験を行うための実験条件を検討している。患者由来肉腫細胞株であるNCC-UPS4-C1細胞(未分化多形肉腫に由来)を用いてスフェロイドを作成した。スフェロイドは三次元に細胞が位置されるため、より生体に近い腫瘍細胞であると考えられている。バイオリアクターと低速のポンプを用いた還流培養を行い、還流培養の前後でのスフェロイドを対象として質量分析(LC-MS/MS)を用いたプロテオーム解析を行った。約2400種類のタンパク質を観察し、とりわけ精度よく比較解析が可能だった1000種類のタンパク質を対象とした解析を行った。その結果、発現が有意に変化するタンパク質を同定することができた。同定した個々のタンパク質については機能解析が行われており、タンパク質の輸送や分解に関係するものや、生体防御に関わるタンパク質が同定された。一方、同定されたタンパク質が多岐にわたるため、それらの変化の統合的な理解は困難だった。スフェロイドを用いた実験系の有用性は確立されたが、その評価にはより網羅性を高めた解析やタンパク質だけでない多層的なオミクス解析が必要であると考えられる。
