Outline of Final Research Achievements |
The mechanism on trafficking of programmed death-ligand(PD-L)1 into cell membrane remains unknown.We discover that BTB binding protein associated with Cullin3(CUL3) E3 ubiquitination ligase is ANKFY1, and that site-specific monoubiquitination of Rab5 by CUL3-ANKFY1 complex regulates the trafficking of PD-L1 into cell membrane.

Academic Significance and Societal Importance of the Research Achievements |
CUL3-ANKFY1 E3ユビキチンリガーゼ複合体の基質の探索研究やAlphascreen systemを用いた ANKFY1-基質結合阻害薬の探索スクリーニングは、これまでにない全く新しい取り組みであり、国内外での報告はない。また、得られた阻害薬によりPD-L1細胞膜輸送を抑制し細胞膜上の発現を減らすことができれば、自己免疫疾患の治療薬につながる可能性もある。本研究で得られた成果は世界をリードするものであり、今後さらなる展開が期待される分野である。
