オレキシンを中心とした内臓知覚過敏の中枢機構解明とその過敏性腸症候群治療への応用

Outline of Annual Research Achievements

一連の研究からオレキシン、グレリン、ドパミン、アデノシン、カンナビノイドやセロトニンによる中枢性内臓知覚鈍麻作用は明らかにし、その網羅的メカニズムをまとめてreview articleにその内容を公表した(Okumura et al., Neuropeptides 2021)。また現在内臓知覚調節の上流に当たる腸管粘膜バリア機能の中枢調節機構を網羅的に解明中で、オレキシンは中枢神経系に作用して迷走神経を介して腸管バリア機能を改善させることが明らかにできた(Okumura et al., Neurosci Lett 2020)。これは腸管バリア機能が中枢性に調節されることを世界で初めて示したもので、ストレスなどで病態が急変する過敏性腸症候群などの病態を考える上で興味ふかい、更にグレリンが中枢神経に作用してオレキシン シグナルを利用して腸管バリア機能を改善することを見出し報告した((Ishioh et al., Neuropharmacology 2020).現在、グレリンの脳内作用メカニズムや新たな神経ペプチドの研究を進めている。

現在の研究をさらに進め、内臓知覚と腸管バリア機能の中枢制御メカニズムの網羅的解析を目指す。
