トランスオミクスが紐解くCOPDの新規バイオマーカーと病態解明

Outline of Annual Research Achievements

トランスクリプトームに関しては、当科においても疾患メカニズムを多くのアレイから同定してきた。肺線維症モデルにおける表現型解析(AJRCCM 2011,AJRCCM2012 読売新聞)だけでなく、CD9 KO mouse(肺気腫感受性)とCD151 KO mouse(肺線維症感受性)のTargetMineによる比較解析から肺気腫と肺線維症の共通・相違遺伝子を同定した(論文受理)。ヒトへの応用へ向けて、既にCOPD患者由来の血清エクソソームmicroRNA/mRNAも解析を進めている。既に申請者がエクソソームから同定したCOPDや喘息における新規BM候補について、疾患特異性、病勢(呼吸機能低下)、急性増悪、予後の観点から当科所有のバンク(3000検体)にて絞り込む。プロテオーム分野では世界トップレベルの技術をもつ専門家(元プロテオーム学会 会長 朝長毅)による定量性と再現性を向上させた次世代プロテオミクス(MRM)を駆使して、大量のBM候補を効率的に絞り込んだ。さらに、COPD患者の肺組織と血清エクソソームのプロテオミクスを比較解析するとともに、マウスモデルでは、ヒトでは回収困難なBAL/肺組織/併存症臓器も経時的に解析する。表現型に最も近いオミクスであるメタボロミクスは、プロテオームやトランスクリプトームといったゲノム情報の媒体の流れの解析では困難な生体反応解明の有望技術と期待されている。既に、申請者は、世界トップレベルの技術を有する福崎教授との共同研究から、肺線維症モデルを用いたメタボロミクスから、新規パスウェイを同定した(論文受理)、次のステップとしてCOPD患者血清にて解析を進行中である。これら種々の階層におけるオミクスデータの統合を、独自のTargetMineにて解析を進めており順調である。
