マスト細胞を標的としたアレルギー疾患根治法の創出に向けたHRFの分子基盤解明

Outline of Annual Research Achievements

ヒスタミン遊離因子(histamine-releasing factor: HRF)がある種のIgE (HRF反応性IgE)と結合し、これを受容体として、このIgEの結合しているマスト細胞を活性化する。しかし、HRFが細胞から分泌される機序の詳細は解明されていなかった。そこで、外界から侵入するアレルゲンに曝されている肺上皮細胞に着目し、HRFの分泌機序について解析し、以下4点を明らかにした。
① アレルゲン、炎症時に産生されるサイトカイン、ATPなどの刺激や物理的な細胞傷害により肺上皮細胞からHRF分泌が誘導される。
② House dust mite (HDM)により誘導されるHRFには、単量体HRF、マスト細胞の活性化能を持つSS結合で形成された2量体に加えて、より高分子量の多量体も存在する。
③ 喘息の増悪因子であるライノウイルス (RV)感染した肺上皮細胞から大量に多量体HRFが分泌される。
④ 定常時の鼻洗浄液には多量体HRFが存在し、RV感染により喘息が悪化した患者ではこの多量体HRF濃度とHRF反応性IgE抗体価が上昇する。
以上より、HRFは、宿主が危険に曝された際にいちはやく周囲の細胞に働きかけ免疫応答を始動させる能力を有する因子(alarmin)であると結論できる。すなわち今まで知られていなかった、アレルギー疾患におけるHRFの新たな機能を提示することができた。さらに、HRFは単量体および2量体のみならず、高分子量の多量体を形成すること、また、喘息の増悪時に多量体HRF量が増加することが示された。
