Outline of Annual Research Achievements

2020年度は2019年度から解析を行っているC. difficileの臨床分離株を用いたagr遺伝子の分子疫学的多様性に関する成果を国際学会や学術誌で報告した。さらにagr遺伝子の機能解明をすすめるため、2020年度よりagr遺伝子欠損株の作成に着手した。C. difficileのゲノム編集には、近年報告されたPolar effectを受けにくいCRISPR-Cas9システムを用いることとした。2020年度にプラスミドベクターの構築を終え、引き続き変異株を作成中である。今後は毒素産生株および非毒素産生株のagr遺伝子の変異株を用い、微生物学的な表現型の検討を行いagr遺伝子の機能解明を行う。
