Outline of Annual Research Achievements

The proposed research strategy involves the following three components (clinical, virologic and immunologic). We have collected 100 serum samples from patients with dengue and severe dengue in Vietnam. Ascites fluid was collected from 40 patients with severe dengue. Inflammatory cytokines were determined by a panel of 10 cytokines to determine inflammatory mediators according to disease severity. Using 8 serum samples and 8 ascites samples, miRNA was isolated and miRNA expression was determined by using microarray. Selected samples will also be analysed by RNAseq, followed by validation by qPCR.

Reason

Dengue patient samples for the characterization of miRNA was been collected together with clinical data. Currently, samples were processed for further analyses by miRNA microarray and RNAseq, as planned in the proposal.

Strategy for Future Research Activity

In the following experiments, levels of antibodies will be determined by using ELISA and neutralizing assay. Because disease outcomes are also affected by sequence of infection, these data will be used to group patients according to disease severity, course of disease and type of infection and analyzed with levels of expressed miRNA. Next, miRNA expression will be determined by RNAseq and microarray by using samples from blood and ascites.
