Outline of Final Research Achievements |
Gene ontology and gene enrichment analyses have revealed that spermatogonia in men with non-obstructive azoospermia are categorized into 4 clusters: 1) Impaired DNA synthesis, 2) impaired receptor expression, 3) impaired stress response and 4)somatic cell type. In men with cluster 1, gonadotoropin treatment to increase spermatogonia DNA synthesis was effective to stimulate spermatogenesis. In cluster 2, aromatase inhibitor may be potentially effective to improve spermatogenesis. Taken together these results, transcriptome analysis using human spermatogonia may open a window toward precision medicine in male infertility.

Academic Significance and Societal Importance of the Research Achievements |
得られた結果は無精子症のみならず一般的な乏精子症や精子無力症の治療にも応用可能と考えられる。究極的には国家的問題である少子化対策に対する基盤研究ともなりえる。単一細胞トランスクリプトーム解析は、精巣内内分泌環境や臨床的背景および精巣の組織学的所見などを統合するといった新しい観点からの解析により、NOAにおける造精機能障害の病態が俯瞰でき、新規薬物療法の開発を見出す手がかりとなった。
