| Outline of Final Research Achievements |
Many institutions choose palatoplasty as a treatment for cleft palate cases, and good results have been obtained. However, palatoplasty has many long-term problems such as maxillary undergrowth caused by postoperative scar contraction, etc., and the development of next-generation cleft palate treatment methods is an urgent issue. In recent years, progress has been made in three-dimensional culture technology including organoids, opening the way to construction of three-dimensional models that enable tissue regeneration closer to natural physiological conditions. In this study, we used amniotic membrane-derived epithelial stem cells as a source and created three-dimensional organoids in vitro using epithelial-mesenchymal interactions based on dental impression-taking molds. , aims at palatal formation closer to physiological tissue.

| Academic Significance and Societal Importance of the Research Achievements |
申請者らは、臨床での口唇口蓋裂治療に応用できないかと考え、口蓋裂隙に充填するオルガノイドを作製し、特に従来の手術療法では回避できない、瘢痕収縮による上顎劣成長、骨露出などの問題点を改善できないかと考えた。 申請者はこれまで口唇口蓋裂に伴う鼻変形治療に対するインプラント型再生軟骨の開発と臨床応用に成功したグループにて再生軟骨の研究に従事しており、特に過去の研究でハイドロゲルと成長因子の組み合わせによる徐放効果を利用した体細胞増殖法を確立している、ハイドロゲルを用いたサイトカインのデリバリーシステムを構築する技術を有している。本研究の遂行により、口唇口蓋裂治療に飛躍的な発展を寄与するものと考えた。
