Outline of Annual Research Achievements |
As for this year, we planned to prepare the matured self-organized tissue rings and evaluate their therapeutic effect on the rat heart with myocardium infarction. Although the project has been affected by the COVID-19, we managed to get the following the achievements: 1.Improved from the matured tissue ring, the 3D tissue sheet paced by the traveling wave have been prepared. The traveling wave could be maintained for over two weeks. This new process allows improved tissue organization and easier transplantation. 2.We proved that the tissue paced by the traveling wave could have better survival rate in hypoxia than the control group. And this could be confirmed by the upregulated expression of metabolism related gene. 3.One paper has been published with the support of grant.

3: Progress in research has been slightly delayed.
Reason
The project has been affected by the pandemic of COVID-19. The lab has been temporally out of service, and some lab members related to this project have to work remotely in home during the most serious phase of pandemic. The order of animals needed for the transplantation were delayed as well.

Strategy for Future Research Activity |
Based on the previous work, we will carry out the following procedures: 1.To transplant the 3D tissue matured by traveling wave to the rat heart with myocardial infarction. 2. To optimize the transplantation condition, such as cell number, tissue size and transplantation timing. 3. To monitor the heart function for over one month post transplantation and to evaluate the retention of transplanted cardiac tissue. 4. To investigate the correlation between the maturation level (such as gene expression, protein expression, contraction motion, electrophysiology and so on) and the retention of the transplanted cells as well as the functional recovery of the infarcted heart.
