Outline of Final Research Achievements |
Anti-brush border antibody disease is a recently reported autoimmune renal disease targeting mainly megalin. It is characterized by a rapid decline in renal functions, leading to end-stage renal failure. However, its prevalence and pathogenesis remain unclear. In this study, we semiquantitatively identified anti-megalin antibodies by western blotting against full-length megalin in patient serum. In addition, we examined individuals clinically suspected of having anti-brush border antibody disease for the presence of autoantibodies. In preparation for enzyme-linked immunosorbent assay (ELISA) for quantification of anti-megalin autoantibodies, a human megalin expression vector was introduced into HEK293 cells to establish a stable human megalin-expressing cell line.

Academic Significance and Societal Importance of the Research Achievements |
抗刷子縁抗体病は世界で20例程度の報告であり、本邦からの報告はなく、現時点では稀な疾患と考えられている。稀であると考えられる一因として、実態把握の調査が行われていないこととともに、診断を確定する簡便な方法が確立されていないことも上げられる。本研究で行われた診断方法の確立に向けての検討によって、今後潜在的な患者の存在が明らかとなる可能性や病因解明の一助にもなる可能性があり学術的意義が大きい。さらに、治療法が確立されれば、将来的な腎代替療法患者を減らす可能性があるとも考えられ、社会的な意義も大きい研究である。
