Outline of Final Research Achievements

Chromosomal Abnormality which frequently occurred in left-sided colorectal cancer was induced to normal karyotypic human iPS cells. Chromosome-induced iPS cells were differentiated into colon in vitro with recombinant protein and inhibitors. Colorectal organoids had digestive-track like structure surrounded by stroma and some part presented with atypia.

Academic Significance and Societal Importance of the Research Achievements

結腸直腸がんでは遺伝子変異のほかに染色体数の異常を、特に原発巣が肛門側に近い症例で高頻度に認める。大腸の発がんやがんの進行に染色体異常がどのように影響しているかを解明することにより、新しい治療法や予防法へとつながる可能性がある。
本研究では、染色体異常をもつ細胞を用いて作成した大腸組織の一部が、正常と比較すると異常と考えられる構造を示していたことから、新たな大腸発がんメカニズムを明らかにできる可能性がある。
