Outline of Final Research Achievements

Adipose tissue plays an important role in the regulation of physiological and pathological metabolism, but little is known about the molecular details. In my previous research, I found that SDF-1, highly expressed in adipose tissue, functions in a cell-autonomous manner, and inhibits insulin sensitivity in adipocytes (Shin et al., Diabetes 2018).In this research proposal, I analyzed the systemic and adipose impact of SDF-1. Adipose-specific knockout of SDF-1 exhibited lower body weight and improved liver steatosis under high-fat diet condition, which was associated with adipose master regulator PPARg and the target genes. the research is under preparation for submission. We also found TGFβ1 functions as cell-autonomous regulator of adipose function. We revealed that the expression of TGFβ1 is increased during feeding, remodels the extracellular matrix, and supports intracellular lipogenesis process in adipocytes (Toyoda and Shin, JBC,2022).

Academic Significance and Societal Importance of the Research Achievements

本研究では脂肪細胞由来分泌因子SDF-1及びTGFβ1が細胞自律的に作用し、脂肪細胞の機能抑制・亢進に寄与することを見出した。これまでSDF-1やTGFβ1は肥満病態を悪化させる炎症性サイトカインとして認識されていたが、その生理的な意義は不明だった。本研究を通し、SDF-1及びTGFβ1の栄養負荷に対してその調整因子としての生理的意義が提唱できた。SDF-1及びTGFβ1の過剰状態では病態を引き起こすことから、今後の肥満及び糖尿病治療に貢献することが期待される。
