Outline of Final Research Achievements

Isotype-specific roles of glutathione peroxidase 4 (GPx4) in the retina had not been clarified. The retina of mitochondrial GPx4 (mGPx4) knockout (KO) mice were investigatigated, and cone-rod dystrophy-like phenotypes were revealed. The degeneration in mGPx4 KO mice were partially rescued by vitamin E supplementation.

Academic Significance and Societal Importance of the Research Achievements

ミトコンドリアは細胞にエネルギーを供給します。ミトコンドリでの脂質の過酸化を防ぐためにglutathione peroxidase 4(GPx4)という抗酸化酵素がありますが、今回の研究ではミトコンドリアにおけるGPx4の発現を消去したマウスの網膜について解析を行い、視細胞という視力や色覚に関わる細胞の細胞死を見出しました。このことによって、GPx4によるミトコンドリアでの過酸化脂質の消去が視細胞にとって必須であることが証明されました。
