Outline of Annual Research Achievements |
The purpose of this project is to investigate the neural mechanisms of Bayesian-like behaviors based on the multi-layer structure of the cerebral cortex. To achieve this purpose, a setup for animal behavior training has been made, and an alternative version of the setup with new features is also being developed. The resting state of neural activities from multiple layers has been taken via a prism lens. The functional connectivity analysis using transfer entropy reveals the the information flow between layers. To address the temporal organization of population neural activities, a hidden Markov model (HMM) is used to extract the hidden brain states. The structure of the function connectivity networks generated by different brain states and metastates is also investigated.

2: Research has progressed on the whole more than it was originally planned.
Reason
This project includes developing a behavior training setup, behavior training, and calcium imaging from multi-cortical layers via a prism probe. I have already finished: 1. Setup: the setup proposed in the proposal has already been made. An alternative design with new features introduced is also under development. 2. Calcium imaging via a prism lens: the surgery for implanting a prism lens (1mm*1mm) to a mouse's brain caused relatively big brain damage. The failure ratio is quite high to get a good field of view, which covers multiple cortical layers, through the prism lens. 3. The resting state of neural activity from multiple layers has been simultaneously taken via a prism lens. Analysis on resting state data reveals the the information flow among cortical layers.

Strategy for Future Research Activity |
1. More behavior training on mice need to be done. The mice need to have prism lens properly implanted, which means with a good field of view on both superficial layers and deep layers. 2. Apply analysis on calcium data taken under training. The analysis methods I have already explored during the analysis on resting state data will be also useful for this analysis, which includes the spike deconvolution methods, and probability density estimation on the deconvolution results. 3. Papers: The analysis on resting state needs to be finished and summarised into a paper. And a possible review paper on existing methods on analysing calcium imaging data is also under preparation.

Causes of Carryover |
1. Experiment cost: the animals(mice), prism lens, and devices for setup (motor and sensors). Due to the low successful ratio of the prism lens implantation surgery, a big amount of mice and prism lens are needed. 2. A high-performance computer is necessary for processing the big calcium imaging data. 3. Travel expense for workshops and conferences
