Outline of Annual Research Achievements |
1. Untargeted metabolomics measurement using HILIC column of the whole cohort has been finished. Data has been processed following the workflow of "retention time correction - data preprocessing in MS-DIAL - manual peak curation - signal correction" and the datasets are ready for further sophisticated data analysis. 2. Untargeted lipidomics analysis platform has been established and data acquisition of the whole cohort in both positive and negative modes has been done. 3. Seventy sphingolipids including ceramides, sphingomyelins,and sphingoid bases etc. were measured targetedly using LC-MS/MS method. This dataset is now being analyzed together with another two asthma cohorts (COPSAC and VDAART) to investigate the role of sphingolipids in different asthmatic cohorts. 4. Data analysis showed that Cortisol in severe asthmatics was significantly lower than that in the controls, indicating that endogenous cortisol might be able to serve as a biomarker of severe asthma. Further investigation should be made to understand the mechanism behind. 5. Pulmonary parameters have been extracted from the biobank and will be incorporated with the datasets including untargeted metabolomics/lipidomics and targeted sphingolipids. 6. Four peer-reviewed papers collaborating with domestic and international researchers have been published this year with the Principle Investigator of this project being listed as a co-author.
