Outline of Annual Research Achievements |
Current project was designed to explore the molecular and neural circuit mechanisms underlying optimistic and pessimistic cognitive bias. In the past fiscal year, we successfully built a behavioral set up to apply ambiguous tone interpretation test on mice. Our data revealed that after training, out-bred CD-1 mice demonstrated 85%successful ratio responding to either positive or negative tones, and there was a considerable individual variation among the subjects in the ambiguous tone. In addition, the retrograding tracing technology and fiber photometry technology was already set up for current project, therefore we can smoothly proceed further to the next step of this project.

2: Research has progressed on the whole more than it was originally planned.
Reason
There are mainly two aims of current project. First, to set up and optimize a behavioral apparatus for analyzing optimistic and pessimistic behaviors of mice. Second, to image the activity of two different populations of cells in BLA during the behavioral analysis. At this stage, we have been successful in setting up the ambiguous-cue interpretation system to check the cognitive bias of mice. We also succeeded in applying retrograde tracing and fiber photometry in our behaving mice. Therefore, we already realized the first aim, and built up the platform to realize the 2nd aim, that is to identify the two populations of cells in BLA and to further image the activity of these two groups of cells during ambiguous-tone interpretation task. We are closer to our goal.

Strategy for Future Research Activity |
By using the behavioral paradigm, tracing and imaging technology built in last fiscal year, we will further current research to the molecular and circuit mechanism level. Basically, we will image the calcium activity of two populations of cells in BLA when mice show positive or negative response during the ambiguous-tone test.

Causes of Carryover |
We will have to purchase and maintain animal, pack virus, and purchase the consumables for calcium imaging analysis(both fiber photometry and mini-scope system). We will also need to attend conference and publish our data.
