Outline of Annual Research Achievements

歯周病原細菌 Porphyromonas gingivalis (以下Pg)の外膜小胞(OMV)の産生様式についての新たな知見を得た。異なる条件で産生されるPg-OMVの物性・組成について調査した結果、Pg-OMVの産生様式として、通常の生育過程で産生される様式と、特定の化学物質がPg菌体に作用して産生誘導される様式の2通りが存在することが明らかとなった。前者と後者のOMVの組成やそ の表面の物理化学的特性は異なっており、特に後者のOMV産生は、Pgの生育に必須の栄養取得の阻害や、その主たる病原因子であるジンジパイン活性阻害の両方に 関連することが明らかとなった。
複数の歯周病原細菌を含む細菌由来OMVがワクチンとして活用できるか、マウスモデルで検討した。Aggregatibacter actinomycetemcomitans(Aa)のOMVについての中枢神経系における安全性と、強力な粘膜アジュバント活性が示された。Pg-OMVの免疫原性はAa-OMVにより増強された。Pg-OMVとAa-OMVを経鼻投与すると、PgとAaの両者に対するワクチン効果(口腔内クリアランスが向上)が認められた。
