Outline of Final Research Achievements

In this study, we developed a kidney-targeted drug delivery system (DDS) using a serine-modified polymer (amino acid cluster) as a new kidney-targeting drug carrier to diagnose and treat renal cell carcinoma. 111In-labeled amino acid cluster predominantly accumulated in the kidney after intravenous injection in mice. In addition, 111In-labeled amino acid cluster hardly distributed to organs other than the kidney, indicating excellent pharmacokinetics with renal selectivity. Tissue distribution imaging using SPECT / CT also showed renal-selective distribution of 111In-labeled amino acid cluster after intravenous injection. Therapeutic radionuclides and drugs-loaded amino acid clusters effectively suppressed tumor growth in the kidney in renal cell carcinoma model mice.

Academic Significance and Societal Importance of the Research Achievements

腎細胞癌の治療効率が低い要因として、腎臓へ薬物が分布しにくいこと、あるいは薬物の腎臓選択性が低いことなどが挙げられることから、腎臓へ効率良く薬物を送達する腎臓指向型ドラッグデリバリーシステム (DDS) の開発が強く望まれる。本研究では、イメージング用・治療用放射性核種などを効率良く腎臓へ送達可能な薬物キャリアを開発することに成功した。これらの知見は腎細胞癌診断・治療法の開発に対して有用な情報を提供するものであると考える。
