Outline of Final Research Achievements

In order to investigate the involvement of glial cells in brain metastasis, I successfully developed a simple and stable culture method MGS (Mixed- glial culture on soft substrate) for prolonged observation of astrocytes and microglia. By using this culture method, I performed drug screening to identify key molecules for brain metastasis-associated glial cells. As a result, I successfully identified a membrane protein X which is important for the cancer promotive phenotype of astrocytes.

Academic Significance and Societal Importance of the Research Achievements

これまでの転移性脳腫瘍研究では適切なin vitroモデルが存在しておらず、マウスを用いたin vivo研究により転移性脳腫瘍の分子メカニズムの解明が行われてきた。そこで、本研究課題で開発したMGS法により、in vitroにおいて脳転移微小環境を模倣することが可能となり、薬剤スクリーニングなどのハイスループットスクリーニングを行うことが可能になった。これにより、極めて予後不良である転移性脳腫瘍の根治療法へとつなげることができると考えられる。
