HSP32を標的とする高分子型制癌剤PEG化亜鉛プロトポルフィリン(ZnP)

Research Abstract

1、水可溶化亜鉛プロトポルフィリン(ZnPP)の高分子型制癌剤として、PEG結合型亜鉛プロトポルフィリン(PEG-ZnPP)およびスチレンマレイン酸コポリマー(SMA)ミセル抱合型ZnPP(SMA-ZnPP)ミセルを合成した。
2、次にPEG-ZnPPおよびSMA-ZnPPの培養腫瘍細胞に対する増殖抑制効果とあわせ、内視鏡Xenon光源光およびLEDの420nmのモノクロマチックの両光照射下での抗腫瘍効果を検討した結果、強い相乗効果がみられた。ヒト白血病CML、細胞K562、ALL、バーキットリンパ腫系、Raji細胞、ATLのMT-2細胞、ヒト食道癌Kyse510、大腸癌SW40、乳癌MCF-7細胞の何れもIC50値が1~10μMであった。
3、In vivoの化学発癌剤DMBA誘発ラット乳癌、マウスS-180およびColon 38に対して、Xenon光(スライドプロジェクター用)および単3乾電池4本のLED光源の420nm附近の光照射(30分)による照射でもPEG-ZnPPおよびSMA-ZnPPの単独i.v.投与に加え、光照射併用群では最も著名な抗腫瘍効果を示した。
4、細胞への取り込みはSMA-ZnPPはPEG-ZnPPの数倍速かった。また、これらはendocytosisにより取り込まれた。PEG-ZnPPのcellへの取り込みが低いことは、いわゆるPEG-dilemmaで説明できる。
5、PEG-ZnPPのアミド結合はセリン型およびチオール型プロテアーゼにより切断され、freeのZnPPの遊離がみられた。
6、SMA-ZnPPミセルから活性本体のZnPPの離脱はレシチンなどの細胞の膜成分の関与でミセルは崩壊した。
