In vivo脊髄抑制性シナプス入力の生理的役割とその可塑的変化の解析

Research Abstract

痛みの伝達に対する抑制性シナプス応答の生理的役割を明らかにするために、以下に焦点を合わせて研究を遂行した。まず、形態学的特徴から脊髄膠様細胞を分類し、樹状突起を頭尾側方向に伸ばすislet細胞が主にGABAなどを含有し、抑制性シナプス伝達に重要な役割を果たしていることが示唆された。また、神経因性疼痛モデル動物を作製して、脊髄の触刺激による抑制機序の変化を解析した。行動実験により触刺激で逃避行動を示すアロディニアが観察されたモデル動物を用いて、抑制性シナプス応答を解析した。しかし、触刺激によって誘起される抑制性シナプス応答はその振幅・発生頻度ともに正常と有意な差は得られなかった。また、痛覚過敏を呈した炎症モデルにおけても抑制性シナプス伝達になんら変化は認められなかった。一方、炎症モデル脊髄膠様質への興奮性シナプス伝達に異常が生じることが明らかになった。即ち、振幅の大きな自発性EPSCの発生、および触刺激に対するEPSCの脱感作が消失するなど過剰な興奮性入力が増大する、さらに、炎症患部に受容野を持つ細胞に特異的にCa^<2+>透過性AMPA受容体を介するEPSCが発生することが見いだされた。この脊髄における炎症患部からの興奮性入力の可塑的変化は、特にC線維の感作によって惹起されることが示唆された。
