神経可塑性に伴うNMDA受容体複合体の再編成と活性化に関与する機能分子の探索

Research Abstract

我々は、神経損傷1週間後でも、グルタミン酸NMDA受容体NR2Bサブユニットの1472番目のTyr残基(Y1472)のリン酸化、神経型一酸化窒素合成酵素(nNOS)の活性化による機能的変化により維持されていること、一酸化窒素(NO)が順行性・逆行性メディエーターが脊髄における神経可塑性に重要な役割をすることを明らかにしてきた。本研究では、神経可塑性に伴うNMDA受容体複合体の再編成と活性化に関与する機能分子の探索するために、
1) 遺伝子改変マウスに慢性疼痛(SNI)モデルを作製し、プロテオミクス解析を行った。野生型無処置群とSNIモデル群、遺伝子改変マウス無処置群とSNIモデル群の4群(各130-140匹)より脊髄後角を摘出し、NMDA受容体複合体が存在するPSD画分精製しプロテオミクス解析を行なった。今年度、4群間の脊髄PSD画分での発現分子の同定、定量化および比較を行い、神経可塑性に伴うNMDA受容体複合体の機能性分子の候補の選定を進めている。
2) NOの下流のカスケードとしてcGMP/sGC/PKG系がよく知られているが、最近、S-ニトロシル化が着目されている。今年度、炎症性疼痛モデルで脊髄後角の蛋白がS-ニトロシル化されること、タンパクのS-ニトロシル化が神経伝達物質の遊離に関与すること、その結果、NOはcGMP/sGC/PKG系を介して一次求心性神経線維の中枢端からのグルタミン酸遊離を促進する一方、抑制系インターニューロンからの遊離を抑制する2面性を有し、脊髄後角での中枢性感作に関与することが示唆された。今回の結果は、ガス状物質NOの痛覚伝達に対する多様性の機序を初めて示したものである。
