Research Abstract

筋細胞や筋組織をマイクロ駆動源材料として適用するために、細胞をアレイ状及びシート状に加工する手法と3次元ゲル構造体として構築する手法を確立し、生命機械システムの試作、原理検証を行った。今年度は連携研究者との共同研究テーマで目標としたデバイスの試作と評価をさらに推進させ、デバイスの機能制御実験を行った。
まず、筋細胞の培養条件の検討、細胞シート化を行うために、連携研究者である大和、秋山ら(東京女子医科大学)の協力を得て細胞シート化する条件の最適化を行い、試作を行った。次に、筋細胞特性による様々なデバイス試作及び微細加工、最適なアセンブリ手法の検討を行った。今年度は特に、様々な形状のマイクロ鋳型を作成することで任意形状の心筋細胞ゲル構造体の作製を行い、異なる大きさ、形状による駆動制御の可能性を示すために評価実験を行った。また、連携研究者である丸尾(横浜国立大学)のマイクロ光造形技術の協力を得て、ゲル構造体とのハイブリッド化を試み、より自由度の高いデバイスを再設計した。マイクロチューブの周囲に培養したチューブ型マイクロポンプの試作、評価を行った。さらに、化学的刺激による筋細胞の制御実験を行い、任意形状に3次元構築したマイクロ心筋細胞ゲルを用いて、収縮力、周波数、変位、寿命の性能評価を行った。さらに、電気刺激や化学刺激などの外部刺激に対するマイクロ心筋細胞ゲルの応答性及び制御性の評価を昨年度に引き続き行い、様々な形状の違いによるマイクロ心筋細胞ゲルの制御、性能向上の可能性を示すことができた。
