Research Abstract

1.フォトクロミック化合物を利用したATPアナログの合成と生体分子モーターとの相互作用
アゾベンゼン誘導体を用いた数種類のATPアナログPABITPの合成に成功した。生体分子モーターであるミオシンとキネシンとの相互作用をATPase活性測定、ミオシン-アクチンあるいはキネシン-微小管の解離実験、エネルギー変換中間体の形成から解析した。このATPアナログは本来のATPと同様に生理的な機能を保持していることが示された。
2.フォートクロミックATPアナログで誘導される分子モーターの構造変化の解析
紫外線と可視光線照射により異なる異性体のフォトクロミックATPアナログで誘導される構造変化を、X線溶液散乱により解析を行った。紫外線-可視光線照射により形成されるATPアナログの二つの異性体はエネルギー変換中間体において、異なる状態を示すことが明らかになった。
3.生体分子モーターの光制御および光駆動実験
キネシン-微小管のin vitro motility assay法を用いてフォトクロミックATPアナログの光異性化により、滑り速度が変化するか実験を行った。紫外線-可視光線照射で可逆的に微小管の滑り速度が変化することが示された。
4.制御系生体分子機械へのフォトクロミック化合物の導入と光制御
低分子量G-protein(Ras)の変異体にフォトクロミック化合物であるアゾベンゼン誘導体を導入して、紫外線-可視光線照射により可逆的にこれらの制御系生体分子機械の機能を制御するための基礎実験を行った。Ras変異体を調製してフォトクロミック分子を導入する実験系を確立することができた。
